Tuff Puffs are an enemy found in the first Paper Mario and come from Huff N. Puff after he is attacked. The more Tuff Puffs there are, the harder it is to cling off Mario and the easier it's for him to take damage. They're also the source of Huff N. Puff, since all he is is a ball of many Tuff Puffs.
